Told him that it's okay u dont want anything more than friendship atleast don't ruin a sis bro bond as he said from his side he see u that way. Also tell him u have to respect our friendship nd ur feeling even he don't like u back it totally fine. Also mention him that don't do these kind of pranks with me don't cross ur boundary.
